摘    要:目的利用高效液相色谱法以芍药苷为指标对清化消食颗粒样品进行定量分析,建立清化消食颗粒中芍药苷的含量测定方法。方法采用高效液相色谱法,色谱柱:Kromasil-C18(4.60mm×250 mm,5μm);流动相:乙腈—水(17∶83);检测波长:230 nm;进样量:10μL;流速:1.0 mL.min-1;柱温:30℃。结果在上述测定条件下,回归方程线性关系良好,回归方程为Y=1 628.634X+28.98,r=0.999 8,n=6,线性范围为0.161~0.805μg,加样回收率为96.2%,其RSD为3.5%。结论本研究建立的HPLC法,有操作方法简捷,分析条件易于精确控制,待测成分易于分离等优点,具有较好的重复性和较高的精密度,可作为清化消食颗粒的含量测定方法。

Abstract:Objective To analyze in quantity the sample of qinghuaxiaoshi granules with HPLC(high performance liquid chromatography)by taking paeoniflorin as the indicator and establish the determination method of paeoniflorin content in the granules.Methods HPLC was adopted,in which chromatographic column:Kromasil C18(250 mm×4.6 mm,5 μm);mobile phase:acetonitrile-H2O(17∶ 83);wavelength:230 nm;sample size:10 μL;flow rate:1.0 mL·min-1 and column temperature:30 ℃.Results The good linear relationship regression equation was obtained with the abovementioned conditions.The regression equation:Y=1 628.634X+28.98,r=0.999 8,n=6;the linear range:0.161~0.805 μg;the average recovery rate:96.2% and RSD:3.5%.Conclusion HPLC method established in this research is simple in operation,in which,the analytic conditions can be accurately controlled and the testing components can be easily separated.This method is advantageous at high repeatability and high accuracy and it can be the one for the content determination of qinghuaxiaoshi granules.
