标准化G422胶质瘤动物模型的肿瘤生长特点

摘    要:目的探讨标准化G422胶质瘤动物模型肿瘤细胞的生长特点。方法将接种G422胶质瘤细胞接种于国际标准化小鼠双侧上肢皮下,每侧分别注射0.1、0.2、0.3ml。接种后5、10、15、20d观察小鼠的生存状态及肿瘤的生长特性;用HE染色及胶质纤维酸性蛋白(GFAP)和S-100蛋白免疫组化分析;AnnexinV-PI观察肿瘤细胞的凋亡情况。结果标准化G422胶质瘤动物模型生长稳定;GFAP及S-100蛋白免疫组化表达强阳性,符合胶质源性肿瘤的特点;肿瘤细胞DNA呈多倍体峰,且早期出现凋亡峰。结论标准化G422胶质瘤动物模型,生长特征及病理表现与人脑胶质母细胞瘤相似,是一种理想的动物模型,为进一步研究提供了重要的平台。

Abstract:Objective To explore the standard rat G422 glioblastoma multiform model.Methods The standard rats(Institute of Cancer Research rat,ICR rat)were inoculated with the tumor of G422 glioblastoma multiform cell line.The ICR rats were injected with 0.1 ml,0.2 ml,0.3 ml tumor liquid.After 5,10,15 and 20 days,the living condition and the character of tumor were observed.Tumor was confirmed with HE and staining of glial fibrillary acidic protein(GFAP)and S-100 protein immunohistochemistry and the apoptosis was observed by Annexin V-PI staining.Results The standard rat G422 glioblastoma multiform model reliant and high rate of tumor production.The GFAP and S-100 protein were obviously positive.The apoptosis was occurred in early stage.All these demonstrated this model resembled histopathological features of human glioma.Conclusions The international ICR rat G422 glioblastoma multiform model resembles histopathological features of human glioma,which can be used as a perfect model to study the etiology of glioblastoma multiform.It could become an important tool for further research.
