摘    要:目的 探讨新生儿T淋巴细胞内信号传递与脐血淋巴细胞凋亡的关系。方法 以健康成人外周血单个核细胞为对照,测定19例足月新生儿脐血淋巴细胞内钙离子浓度,蛋白激酶A活性和蛋白激酶激动剂PMA对脐血淋巴细胞凋亡的影响。结果 (1)经抗-CD3刺激前或刺激事脐血单个核细胞和PBMC细胞内钙离子浓度差异无显著意义;(2)PMA可纠正抗CD3诱导的CBMC凋亡;(30培养2小时,CBMC PKA活性自发显著升高

Abstract:Objective In order to explore the effect of intracellular signal transduction on apoptosis of cord blood mononuclear cells. Methods Intracellular calcium concentration, protein kinase A (PAK) activity, and the effect of PMA, which is the activator of protein kinase C (PKC) on apoptosis of cord blood mononuclear cells (CBMC) were determined in CBMC with the control of normal adult peripheral blood mononuclear cells (PBMC). Results There were no differences in calcium concentrations between CBMC and PBMC before or after stimulation of antiCD3. PMA could completely correct the CBMC apoptosis induced by antiCD3. Spontanous PKA activities in CBMC cultured for 2 hours in absence of mitogen increased from 1.99 to 2.37 (P<0.01). While PKA activities in PBMC decreased. Conclusion The imbalance of intracellular signal transduction might be the main cause of apoptosis induced by antiCD3 in CBMC. But spontanous apoptosis in CBMC might related to increased PKA activities.
