ET-1、MMP-3、TIMP-1在人肾小球中的表达及意义

摘    要:目的 研究内皮素 1(Endothelin 1,ET 1)、基质金属蛋白酶 3 (Matrixmetalloprotevnase 3 ,MMP 3 )、金属蛋白酶组织抑制剂 1(Tissueinhibitorofmelalloproteinase ,TIMP 1)在人肾小球中的表达变化及意义。方法 应用免疫组化技术检测3 2例原发性系膜增生性肾炎患者及 5例对照组肾小球中ET 1、MMP 3、TIMP 1、LN蛋白的表达。结果 在中、重度系膜增生性肾炎患者的肾小球中 ,ET 1、MMP 3、TIMP 1、LN蛋白表达增强 ,MMP 3 /TIMP 1比值下降 (P <0 0 5 ,P <0 0 1) ;ET 1的表达与MMP 3 /TIMP 1比值负相关 (r =-0 70 3 ,P <0 0 1) ,与LN表达正相关 (r =0 880 ,P <0 0 1) ;MMP 3 /TIMP 1比值与LN表达负相关 (r=-0 62 3 ,P <0 0 1)。结论 ET 1可能是肾小球MMP 3 /TIMP 1下降的原因之一 ,这与肾小球细胞外基质 (Exlracellularmatrix ,ECM)降解受抑密切相关。

Abstract:Objective To investigate the expressions of endothelin 1(ET 1), matrix metalloproteinase 3(MMP 3) and tissue inhibitor of metalloproteinase 1(TIMP 1) in human glomeruli and the related significance. Methods A total of 32 patients with primary mesangioproliferative glomerulonephritis(MsPGN) and 5 controls were included in this study. The expression levels of ET 1, MMP 3, TIMP 1 and LN proteins were determined by immunohistochemistry. Results The expressions of ET 1, MMP 3,TIMP 1 and LN proteins enhanced in normal glomeruli of moderate and severe MsPGN. The ratio of MMP 3/TIMP 1 decreased in severe MsPGN( P <0.05 or P <0.01). The expression of ET 1 was negatively correlated with the ratio of MMP 3/TIMP 1( r =-0.703, P <0.01) but positively with the expression of LN( r =0.880, P <0.01). The ratio of MMP 3/TIMP 1 was negatively correlated with the expression of LN( r =-0.623, P <0.01). Conclusion ET 1 may be involved in the decrease of the ratio of MMP 3/TIMP 1, which may contribute to the inhibition of ECM degradation in human glomeruli.
