山东半夏不同炮制品相关毒性物质基础含量与急毒实验比较研究

摘    要:目的进行山东半夏不同炮制品相关毒性物质基础含量与急性毒性的比较研究,考察炮制对相关毒性物质基础含量与毒性的影响。方法运用UV法、HPLC法、酸碱滴定法和经典急性毒性实验法,进行半夏、清半夏水提组分总生物碱、总有机酸及其可测单一成分的含量测定和急性毒性比较研究。结果相关物质基础含量测定结果显示:总有机酸、总生物碱和鸟苷含量大小为半夏〉清半夏;琥珀酸、麻黄碱含量为清半夏〉半夏。毒性研究结果显示:半夏、清半夏水提组分MLD值为301.2g/kg和374.4g/kg,分别相当于临床70kg成人每公斤体重日用量的2342.7倍和2912.0倍。结论炮制对山东半夏相关毒性物质含量与毒性有一定的影响,半夏毒性大于其炮制品清半夏。

Abstract:Objective To compare the content of toxical substances and the acute toxicity of Shandong Pinelliae's different extractions,and discuss the extraction's influences on the content of toxical substances and acute toxicity.Methods UV method,HPLC method,acid-base titration and the classical method of acute toxicity were used to determine the content of toxical substances and compare the acute toxicity of Pinelliae and its extraction.Results The content of total organic acid,total alkaloid and vernine is:Pinelliae extraction;the content of amber acid and ephedrine is: extraction Rhizoma Pinelliae.The MLD of Pinelliae is 301.2g/kg,and its extraction is 374.4g/kg,which is respectively equal to 2 342.7 times,2 912.0 times of 70kg people's daily dried medicinal herb expenses.Conclusion Extraction has certain influence on the content of toxical substances and acute toxicity of Rhizoma Pinelliae,and the toxicity of Rhizoma Pinelliae is higher than its extraction.
