蒺藜皂苷预适应对大鼠心肌缺血/再灌注损伤的保护作用

摘    要:目的探讨蒺藜皂苷(GSTT)预适应对大鼠心肌缺血/再灌注损伤的保护作用。方法 56只大鼠随机分假手术组(Sham)、缺血/再灌注(I/R)组、缺血预适应组(IP)、阳性药(尼可地尔1.0mg·kg-1)组、GSTT100、30、10mg·kg-1组。采用分光光度法测定血清乳酸脱氢酶(LDH),二氨基二苯甲烷(MDA),超氧化物歧化酶(SOD)水平。ELISA法测定血清TNF-α与IL-6的含量。光学显微镜下观察受损心肌病理组织学改变,采用TUNEL法检测心肌细胞凋亡,Western blot检测心肌组织内caspase-3表达。结果与模型组比较,GSTT100、30mg·kg-1组LDH、MDA含量明显降低,SOD活性明显升高(P<0.01),TNF-α及IL-6含量也明显降低,凋亡细胞数明显减少(P<0.01),caspase-3表达下降,心肌组织形态明显改善。结论蒺藜皂苷预先给药对大鼠心肌缺血/再灌注损伤具有保护作用,能减少自由基生成,抑制细胞凋亡。

Abstract:Aim To explore the gross saponins of Tribulus terrestris(GSTT)preconditioning on myocardial ischemia-reperfusion injury in rats.Methods The animal model of acute myocardial ischemic and reperfusion was established.Rats were randomly divided into sham operation group,ischemia/reperfusion group,ischemic preconditioning group,nicorandil 1.0 mg·kg-1 group,and GSTT 100,30,10 mg·kg-1 groups.Lactate dehydrogenase(LDH),2-amino diphenyl methane(MDA),and superoxide dismutase(SOD)levels were assessed and evaluated by spectrophotometry.The serum TNF-α and IL-6 levels were evaluated by ELISA.The histopathological changes of myocardial damage was observed by optical microscope.Apoptosis in myocardial tissue was detected by TUNEL.Results Compared with model group,the contents of LDH,MDA in GSTT 100,30 mg·kg-1 groups were lower,SOD activity was significantly higher(P<0.01),and TNF-α and IL-6 levels also decreased significantly.The number of apoptotic cells decreased(P<0.01)and myocardial morphology improved significantly in GSTT group.Compared to the I/R group,caspase-3 was significantly lower in GSTT groups.Conclusion GSTT pretreatment has a protective effect on myocardial ischemia-reperfusion injury by inhibiting cell apoptosis,which involves the reducing of free radicals.
