Abstract:Objective: To investigate the inhibition and apoptosis-induction effect of combination of Nimesulide and adriamycin on osteosarcoma MG-63 cells.Methods: MG-63 cells were treated with different concentrations of Nimesulide and adriamycin.Cell growth was analyzed with MTT assay.Cell cycle and apoptosis were tested by flow cytometry.Expression of β-galactosidase,a senescence induced marker,was analyzed by the β-Gal assay.Cell cycle related proteins were tested by Western blot.Results: The combination of Nimesulide and adriamycin inhibited the growth of MG-63 cells more effectively than adriamycin and in a concentration-time dependent way.The combination of adriamycin and lower concentration of Nimesulide induced a dose-dependent increase in G2/M phase arrest and apoptosis;while the combination of adriamycin and higher Nimesulide induced a senescence-like G0/G1 phase arrest,along with up-regulated expression of bax and p21 and down-regulated expression of bcl-2,cyclin E,C-myc,and CDK2.Conclusion: Cell growth inhibition rate was significantly increased by combination of Nimesulide and adriamycin,which provided the theoretical basis for clinical combination application.
