胰岛素对小鼠早期胚胎体外发育的影响

摘    要:目的 探讨胰岛素对小鼠体内外来源早期胚胎体外发育的影响。 方法 用添加不同浓度胰岛素和胰岛素+葡萄糖的CZB培养液对小鼠体内外来源胚胎进行体外培养 ,观察囊胚发育率和囊胚细胞数的变化。 结果 小鼠 2 细胞胚胎体外培养 10 2h后 ,0 .0 5 μg/ml胰岛素添加组扩张囊胚率、孵化囊胚率和囊胚细胞数均显著高于其他各浓度添加组。葡萄糖加入显著提高了这一效应。添加 0 .0 5 μg/ml胰岛素和葡萄糖的CZB培养液同样显著提高体外受精 (IVF)胚胎的孵化囊胚率和囊胚细胞数。 结论 添加胰岛素和葡萄糖培养液对体内或IVF小鼠胚胎发育具有相同的促进作用。

Abstract:Objective: To evaluate the influence of insulin on the development of mouse preimplantation embryo in vitro. Methods:Two-cell mouse embryos, collected from uterine flushing or IVF, were cultuted for 102 hours in the CZB media supplemented with insulin at different concentrations or with insulin and glucose. The rate of development to blastocyst and the cell number of each blastocyst were determined. Results: Insulin at the concentration of 0.05 ng/ml caused a significant increase in the rate of expanded blastocyst, hatched blastocyst and the cell number of blastocyst, while the glucose enhanced this promoting effect from insulin. Conclusion: Insulin can promote the in vitro development of mouse preimplantation embryo by means of stimulating the cell differentiation and glucose metabolism.
