摘    要:目的研究碱性成纤维细胞生长因子(bFGF)对新生大鼠缺氧缺血脑损伤(HIBD)后血管内皮生长因子(VEGF)表达的影响及其意义。方法72只新生7日龄SD大鼠分为三组,每组24只,(1)假手术组(2)缺氧缺血组(3)bFGF干预组。制备大鼠HIBD模型后分别于即刻、3小时、12小时、24小时、3天、7天将动物处死(N=4)。采用SABC免疫组化方法检测VEGF蛋白的表达。结果假手术组海马和皮质VEGF不表达。VEGF蛋白在大鼠缺氧缺血脑损伤后的表达增强,即刻有增加,12小时达高峰持续至24小时后下降。经bFGF干预后VEGF蛋白表达较缺氧缺血组增高,VEGF蛋白表达bFGF干预组与缺氧缺血组相比3小时有统计学差异,持续至3天(p<0.05)。结论外源性bFGF能明显增加新生大鼠缺氧缺血后脑组织VEGF表达,在缺氧缺血脑损伤再生与修复中发挥一定保护作用。

Abstract:Objective To explore influence and significance of exogenous bFGF on VEGF expression in hypoxia-ischemia brain of neonatal SD rat.Methods Seventy-two SD rats of 7-day-old were randomly assigned into a sham-operation group,a HIBD group and a bFGF intervention group(n=24 each).Each group was randomly subdivided into subgroups sacrificed immediately,at 3,12and 24 hrs and 3 and 7 days after HI(n=4).Expressions of VEGF were examined with immunohistochemical staining and image quantitative analysis.Results VEGF was not expressed in the hippocampus and cortex in the sham-operated group.It is obviously that the number of VEGF positive cells in the hippocampus and cortex increased reactively after HI compared with the control group(p<0.05) and reached peak 12 hours and persistent to 24 hours.Exogenous bFGF enhanced the expression of VEGF in hippocampus and cortex of the neonatal rat 3 hours to 3 days after ischemia and hypoxia(p<0.05).Conclusion Exogenous bFGF can up-regulate the VEGF expression in neonate rats following HIBD,which may play important roles in restoration of neurons damaged by hypoxia-ischemia.
