摘    要:目的以存档蜡块,选择最佳抗原修复液,而进行抗原修复,使抗原决定簇暴露,达到最理想染色结果。方法用柠檬酸缓冲液(pH6.0)、EDTA缓冲液(pH9.0),及高胝加热法,胰蛋白酶消化对CD68、lysozyme、α—AT和S-100不同的抗体进行修复。消化,规合抗原决定簇暴露。,结果3种抗原修复法的免疫组化切片,胰蛋白酶修复法阳性率显著高于高胝加热法与微波加热法且染色背景清晰,高压加热法与微波加热法非特异性着色较深,阳性率低。结论对不同的抗体选用不同的修复方法可以达到最佳效果.

Abstract:Objective To explore the best reeovery modes of some antibodies for immunohistochemical stain- ing. Methods The antibodies, as CD68, lysozyme, α-AAT and S-100, were repaired and digested separately by high-pressure heating, trypsin digestion and microwave heating. Meanwhile, their species specific epitopes exposure and staining effect were observed. Results In three species antigen recovery methods of immunohisto- chemical sections, the positive rate of trypsin recovery method was significantly higher than those of high-pressure heating and microwave heating. The staining background under microscope showed also clear in trypsin recovery method, and with dark non-specific staining in high heating and microwave heating. Conclusion The adoption of different recovery modes to different antibodies can obtain the best staining effect.
