miR-769对食管鳞癌细胞增殖、侵袭和迁移能力及紫杉醇耐药性的影响

摘    要:目的:探讨hsa-miR-769(miR-769)在食管鳞癌组织中的表达及其对癌细胞增殖、侵袭和迁移能力及紫杉醇耐药性的影响。方法:通过生物信息学技术挖掘,我们对肿瘤相关的hsa-miR-769(miR-769)的表达进行了深入研究,进而进行临床样本的验证,并通过细胞生物学和分子生物学研究对其作用机制进行了进一步探索。结果:公共数据库的挖掘结果显示miR-769在食管癌组织中表达异常升高,且在临床样本中我们观察到了相同的升高异常,通过细胞实验和分子生物学实验,我们发现miR-769调控肿瘤细胞的增殖、侵袭和迁移能力,同时引起肿瘤细胞的上皮间质转化过程发生改变,后续的研究中,我们成功构建了紫杉醇耐药的食管鳞癌细胞系,并发现miR-769在食管鳞癌对于紫杉醇的获得性耐药中起到了重要作用。结论:miR-769影响食管鳞癌细胞的增殖、侵袭和迁移能力及对紫杉醇的耐药性,本研究旨在探索食管鳞癌发生发展的机制,并为食管鳞癌靶向治疗提供了新的潜在靶点。

Abstract:Objective:To investigate the hsa-miR-769(miR-769) expression in esophageal squamous cell carcinoma and its effect on the proliferation,invasion,migration and paclitaxel resistance of cancer cells.Methods:Through bioinformatics technology mining,we conducted an in-depth study on the expression of tumor-associated hsa-miR-769(miR-769),and further verified clinical samples,and further explored its mechanism of action through cell biology and molecular biology studies.Results:The mining results of public database showed that miR-769 expression was abnormally elevated in esophageal carcinoma tissues,and the same abnormal expression was observed in clinical samples.Through cell experiments and molecular biology experiments,we found that miR-769 regulated the proliferation of tumor cells invasion and migration ability can also cause changes in the epithelial mesenchymal transformation process of tumor cells.In subsequent studies,we successfully constructed a paclitaxel-resistant esophageal squamous cell carcinoma line,and found that miR-769 played an important role in the acquired resistance of esophageal squamous cell carcinoma to paclitaxel.Conclusion:miR-769 affect the proliferation,invasion,migration and resistance to paclitaxel of esophageal squamous cell carcinoma.This study aims to explore the mechanism of the occurrence and development of esophageal squamous cell carcinoma,and provide a new potential target for targeted therapy of esophageal squamous cell carcinoma.
