芦荟大黄素纳米脂质体联合5-氟尿嘧啶对人舌鳞癌CAL-27细胞增殖、凋亡、侵袭及迁移的影响

摘    要:目的:探究芦荟大黄素纳米脂质体(aloe-emodin nanoliposome,AE-L)联合5-氟尿嘧啶(5-fluorouracil,5-FU)对人舌鳞癌CAL-27细胞的作用效果。方法:逆相蒸发法制备AE-L;CCK-8检测应用不同浓度AE-L和5-FU对CAL-27细胞抑制率的影响;克隆形成实验检测细胞增殖;划痕修复实验检测细胞迁移能力;Transwell侵袭实验检测细胞侵袭能力;Hoechst 33342检测细胞凋亡情况;实时定量荧光PCR(real-time quantitative fluorescent PCR,qRT-PCR)检测Bcl-2、Caspase-3 mRNA的表达情况。结果:芦荟大黄素纳米脂质体联合5-FU能够明显抑制CAL-27细胞的增殖,降低细胞迁移能力,同时还可以降低细胞侵袭能力。通过Hoechst 33342染色可发现联合用药组细胞凋亡数量明显高于空白对照组和单药组。qRT-PCR检测可发现联合组抗凋亡基因Bcl-2表达明显下降,凋亡基因Caspase-3表达明显增高。结论:芦荟大黄素纳米脂质体联合5-FU对人舌鳞癌CAL-27细胞具有明显的抑制作用,其机制可能与促进凋亡发生相关。

Abstract:Objective:To investigate the effect of aloe-emodin nanoliposome(AE-L) combined with 5-fluorouracil(5-FU) on human tongue squamous cell carcinoma CAL-27 cells.Methods:AE-L was prepared by reverse-phase evaporation.CCK-8 was used to detect the effect of different concentrations of AE-L and 5-FU on the inhibition rate of CAL-27 cells.Clone formation assay was used to detect cell proliferation.Scratch repair assay was used to detect cell migration ability.Cell invasion ability was detected by Transwell invasion assay.Hoechst 33342 was used to detect cell apoptosis.The expression of Bcl-2 and Caspase-3 mRNA was detected by Real-time fluorescent quantitative PCR(qRT-PCR ).Results:AE-L combined with 5-FU can significantly inhibit the proliferation of CAL-27 cells,reduce cell migration ability,and also reduce cell invasion ability.Hoechst 33342 staining showed that the number of apoptotic cells in combination group was significantly higher than that in blank control group and single drug group.The expression of anti-apoptotic gene Bcl-2 was significantly decreased and the expression of apoptotic gene Caspase-3 was significantly increased by qRT-PCR in combination group.Conclusion:AE-L combined with 5-FU inhibit human tongue squamous cell carcinoma CAL-27 cells,and its mechanism may be associated with apoptosis.
