免疫荧光技术在生物材料体外细胞培养实验中的应用

摘    要:作者根据荧光显微镜的成像原理,采用免疫荧光染色技术将钛片表面成骨细胞骨架-肌动蛋白特异性荧光标记,吧呈现细胞的形态大小以及细胞核。结果表明:束状排列的荧光着色向一丝位于胞 浆内,并勾画出细胞外形和细胞核的轮廓。

THE APPLICATION OF IMMUNOFLUORESCENT TECHNIQUE IN BIOMATERIAL CYTOLOGICAL RESEARCH
Abstract:Based on the working mechanism of fluorescent microscope, the authors specificly labelled the actin microfilament of osteoblast with fluorescein by indirect immunofluorescent staining method to show the morphologies, sizes and the nuclei of the cells cultured on Ti plates. The results showed that the cells were composed of a bundle of microfilaments labelled with fluorescein in the cytoplasm, which presented the outlines of the nuclei as well as of the cells. The shapes of the cells cultured on Ti plates were mainly triangular or polygonal, which are in accordance with the results by SEM.The authors hold that, besides the shapes,the immunofluorescent technique can also molecularly show the characteristics and the functions of cells cultured on biomaterials,and thus is better than the SEM technique to some extent in this area.
