摘    要: 目的:建立测定人血浆丙泊酚(异丙酚)浓度的HPLC方法,为临床异丙酚的药理学研究提供科学的分析技术?方法:用环己烷提取血浆中的异丙酚,用HPLC测定异丙酚含量?测定异丙酚以μ-Bondapak C18柱为分离柱,乙腈-水(55∶45)为流动相,以百里酚为内标物,用紫外检测,波长270nm?结果:本方法回收率高,重现性好,线性范围为0.1~4.0μg·ml-1,最低检测浓度为0.05μg·ml-1?结论:本方法灵敏、快速、选择性强,具有较高的精密度和准确度,适用于临床血药浓度检测?

Abstract:OBJECTIVE: To develop a method for the determination of propofol concentration in plasma by using HPLC and to provide scientific analysis for the studies of the pharmacology of propofol in clinical anesthesis.METHOD: The propofol in plasma was extracted with cyclohexane and was determined by HPLC.The propofol was separated on μ-Bondapak C18 column with a mobile phase of acetonitril-water (55∶45) and detected at 270 nm.Thymol was used as the internal standard.RESULTS: This method was high recovery and fine reproducibility.The linear range were 0.1~4.0 μg·ml-1.The limit of detection was 0.05 μg·ml-1.CONCLUSION: The reversed-phase HPLC described in this report is rapid, precise, accurate and can be used to measue propofol plasma levels in clinic.
