三七总皂苷对免疫性肝损伤小鼠的保护作用

摘    要:目的研究三七总皂苷对免疫性肝损伤小鼠的保护作用。方法采用BCG2.5 mg静脉注射致敏小鼠,第10天给于LPS 7.5μg静脉攻击造成小鼠免疫性肝损伤模型,灌胃(ig)给予PNS(100、200、400 mg/kg)连续10 d。进行肝、脾脏器指数的测定;分光光度法测定血清ALT、AST浓度以及肝匀浆中MDA、SOD、GSH-Px的含量;放射免疫分析肝脏肿瘤坏死因子-α(TNF-α)、白细胞介素1β(IL-1β);HE染色法作肝组织病理学检查;免疫组织化学研究TGF-β1、核因子(NF)-κB p65的表达;RT-PCR法检测肝脏组织中TNF-amRNA表达。结果免疫性肝损伤小鼠血清ALT、AST较正常组明显升高,肝脏中MDA明显升高而SOD、GSH-Px较正常组明显降低。PNS灌胃能显著降低免疫性肝损伤小鼠肝脏、脾脏指数(P<0.05,P<0.01),降低血清中升高的ALT、AST含量,降低肝匀浆中MDA水平,同时升高其低下的SOD、GSH-Px水平;抑制TNF-α、IL-1β水平;HE染色法肝组织病理学检查显示PNS能改善肝组织病理损伤;PNS也能降低免疫性肝损伤小鼠TGF-β1和NF-κBp65在肝脏中的强烈表达;RT-PCR检测肝脏组织中TNF-amRNA表达免疫性肝损伤有明显的抑制作用。结论三七总皂苷对免疫性肝损伤小鼠具有保护作用。

Abstract:Objective To observe the protective effects of panax notoginseng saponins on immunological liver injury in mice.Methods The model of mouse immunological liver injury was induced by injecting bacillus calmette guein(BCG) and lipopolysaccharide(LPS) into tail vein.Visceral parameters of liver and spleen were determined;levels of ALT and AST in serum,the activity of SOD,GSH-Px and the of MDA concentration were assayed by spectrophotometry;the levels of TNF-α and IL-1β were detected by ELISA;the coefficient of liver were assayed and dimensions of hepatic damage in the histopathology were observed;TGF-β1 and NF-κB p65 in hepatic tissues were determined by the method of immunohistochemistry;Liver samples were collected after the experiment and stained by immunohisfochemistry of TNF-amRNA.Results PNS was found to significantly decrease coefficients of liver spleen(P<0.05,P<0.01),also decrease the serum transaminase activity,MDA and prevent the reduction of SOD and GSH-Px activities in liver homogenate and also effectively prevent the levels of TNF-α and IL-1β;histopathological observations also showed that PNS could ameliorate the liver injury in different degrees and were correlated with the biochemical parameters and that PNS could effectively prevent the expressions of TNF-β1 and NF-κB p65;moreover PNS significantly reduced TNF-amRNA in liver.Conclusion PNS has protective effects on immunological liver injury in mice.
