人胚脾LAK细胞体外抗肿瘤作用的实验研究

本文对人胚脾ＬＡＫ细胞的制备及其体外抗肿瘤作用进行了实验研究，为临床推广应用提供了可行性的依据。实验表明，在红细胞低渗休克法，溶血盐法，淋巴细胞分离液分离法中，以最后一种方法制备人胚脾ＬＡＫ前体细胞的效果最佳。用重组白细胞介素２诱导的人胚脾ＬＡＫ细胞在培养的第５～８ｄ对Ｋ５６２靶细胞的细胞毒活性最高，达６５．１％；而对Ｒａｊｉ靶细胞的细胞毒活性较低，在培养的第５ｄ为２６．７％。由此可见，人胚脾ＬＡＫ细胞对Ｋ５６２和Ｒａｊｉ两种肿瘤细胞均有杀伤活性。

An Experimental Study on Anti-tumor Effect of HFS-LAK Cells in vitro

In this paper, preparation and anti-tumor effect in vitro of HFS-LAK cells were studied experimentally,providing possible evidence for clinical application extensively. The results showed that, among red blood cell hypotonic shock techniqne etc,the last technique with which the pre-HFS-LAK cells being prepared was the best. Cytotoxicity of HFS-LAK cell activated with rIL-2 against k562 was highest on the days of culture from 5th to 8th, reaching 65.1%,and that against Raji was lower, only showing 26.7%on the 5th day of culture.Therefore HFS-LAK cells could have cytotoxicity against both K562 tumor cells and Roji tumor cells.
