姜黄素对人鼻咽癌细胞CNE-2Z细胞骨架的影响

摘    要:目的:观察姜黄素对鼻咽癌细胞CNE-2Z细胞骨架的影响,探讨姜黄素对鼻咽癌细胞直接杀伤作用的可能机制。方法:用考马斯蓝染色和图像分析方法检测不同浓度姜黄素作用24 h后CNE-2Z细胞骨架的染色变化;用免疫组织化学方法观察表皮生长因子受体(EGFR)、增殖细胞核抗原(PCNA)、磷脂酰肌醇3激酶(PI3K)、Tubulin-β在各组细胞中的表达情况;运用RT-PCR方法检测不同浓度姜黄素作用后的CNE-2Z细胞中EGER的表达水平。结果:CNE-2Z细胞在10μmol/L姜黄素处理后细胞体积有所增大,随后姜黄素浓度升高细胞骨架染色逐渐降低(P<0.01);不同浓度的姜黄素可抑制CNE-2Z细胞中EGFR、PCNA、PI3K和Tubulin-β的表达,并使EGFR mRNA在CNE-2Z细胞中的表达下调。结论:姜黄素可能通过下调EGFR影响细胞骨架,从而起到直接杀伤鼻咽癌细胞CNE-2Z的作用。

Abstract:Objective: To observe the effects of curcumin on the cystoskeleton of nasopharyngeal carcinoma cell CNE-2Z,and to approach the possible mechanisms of curcumin on direct lethal effect of nasopharyngeal carcinoma cell.Methods: After 24 hours' treatment with curcumin,Coomassie blue staining and image analysis were used to measure the cystoskeleton of CNE-2Z cells.Immunohistochemistry was used to observe the expression of EGFR,PCNA,PI3K,and Tubulin-β.RT-PCR was performed to detect EGFR mRNA of CNE-2Z cells after 24 hours' treatment with curcumin at different concentration.Results: CNE-2Z cells were in bigger size after treated with of curcumin at 10 μmol/L,then cystoskeleton staining became stronger following the treatment with higher concentration of curcumin(P<0.01).The curcumin at different concentration inhibited the expression of EGFR,PCNA,PI3K,and Tubulin-β,along with a down-regulation of EGFR mRNA in CNE-2Z cells.Conclusion: Direct lethal effect of nasopharyngeal carcinoma cell by curcumin was possibly associated with the influence of cystoskeleton induced by down-regulation of EGFR.
