三七总皂甙对肺缺血再灌注时环氧酶-2及其基因表达的影响

摘    要:目的:探讨三七总皂甙(panax notoginseng saponins,PNS)对肺缺血再灌注时环氧化酶表达的影响.方法:30只日本大耳兔均分为三组:假手术组(S组),缺血再灌注组(IR组)和缺血再灌注 三七总皂甙(PNS组).检测血清丙二醛(MDA)含量、超氧化物歧化酶(SOD)和黄嘌呤氧化酶(XO)活力,肺组织环氧酶-1(COX-1)、环氧酶-2(COX-2)蛋白表达;肺组织COX-1mRNA、COX-2 mRNA表达.结果:PNS组血清MDA、XO均显著低于IR组,SOD明显高于IR组(P<0.05和P<0.01);PNS组肺组织COX-2蛋白和COX-2mRNA表达皆显著低于IR组(均P<0.01),肺组织COX-1蛋白和COX-1 mRNA表达三组间无明显变化.结论:PNS可通过增强抗氧化应激和下调肺组织COX-2蛋白及其基因的表达而减轻肺缺血再灌注损伤.

Abstract:Objective:To observe protective effects of panax notoginseng saponins (PNS) during lung ischemia reperfusion injury (LIRI) and investigate its mechanism. Methods: Rabbit lung model of ischemia reperfusion injury was constituted in vivo. Thirty rabbits were divided into three groups randomly: sham operation group(S group), ischemia reperfusion group(IR group) and ischemia reperfusion plus panax notoginseng saponins (PNS group ). Malondialdehyde (MDA), superoxide dismutase (SOD) and xanthine oxidase (XO) in serum were measured. The expressions of COX-1 and COX-2 protein were measured in lung tissue by immunohistochemistry (IHC).The expressions of COX-1mRNA and COX-2mRNA were observed in lung tissue by in situ hybridization (ISH). Results: In IR group, XO and MDA increased and SOD decreased in serum, while the same changes happened in PNS group but less severely.(P<0.01). The IHC and ISH demonstrated that COX- 2 protein and COX-2mRNA in pulmonary tissue of PNS group were significantly lower than those of IR group (P<0.01). The difference of COX-1 protein and COX-1mRNA in pulmonary tissue was not significant in three groups. Conclusion: Panax notoginseng saponins may attenuate lung ischemia reperfusion injury through oxidation resisting process to reduce the COX-2 in lung tissue and inhibit cyclooxygenase-2 expression.
