摘    要:目的运用ICM(Im age Cytom eter)对图像进行处理和分析,在给出细胞形态学参数的同时对DNA的含量进行定量分析,能有效地协助医生对诸如肿瘤等多种病症做出诊断。方法以武汉大学光谱与成像仪器工程技术研究中心研制的“全自动DNA细胞图像定量分析仪”(DNA-ICM)为平台,对鸡血红细胞和宫颈脱落细胞进行液基薄层制片,用Feu lgen染色法染色,对DNA-ICM上获取的细胞染色图像进行扣背底算法处理。结果用冷酸解法对两种细胞进行Feu lgen染色后细胞着色较深且均匀;进行扣背底算法处理后的细胞图像的灰度值方差为0.524,比不进行处理时(0.919)要小。结论冷酸解法更适用于鸡血红细胞和宫颈脱落细胞的Feu lgen染色;扣背底算法可以有效地减少视场不匀带来的误差。

Abstract:ObjectiveImage processing and analyzing by use of ICM(Image Cytometer)can be conductive to doctors in tumor diagnoses.MethodsMonolayer of chicken red blood cells and cast-off cervical cells were deposited onto microscopic slides by liquid based sampling preparation.The slides were stained by Feulgen coloration for determination of amount of DNA using an automated DNA imaging cytometer(made by Engineering Research Center of Spectrum & Imaging Instrument,Wuhan University) and cell images obtained were processed by the method of distracting background in image processing.ResultsThe color of two types of cells after cold acidolysis used in Feulgen coloration was deeper and more symmetrical than that after hot acidolysis.The gray scale variance of cell images after disposure of distracting background was smaller than that of no such disposure.ConclusionCold acidolysis was more suitable in Feulgen coloration of chicken red blood cells and cast-off cervical cells.Method of distracting background could effectively diminish the asymmetry of the background of cell images.
