Abstract:Objective:To study the mechanism of bisphosphonates in the prevention and treatment of aseptic loosening of prosthesis on cytokines-related genes modulation.Methods:In vitro human peripheral blood mononuclear cells(PBMC) were cultured with wear particles and sodium allendronate of different concentration.Then expression of IL-6m RNA was determined by In Situ Hybridization and RT-PCR techniques.Results:The expression of IL-6mRNA was increased significantly when wear particles was added as compared with control group ( P <0.01).Hybridization and RT-PCR demonstrated that bisphosphonates reduced IL-6mRNA expression.Conclusion:Wear debris can stimulate IL-6mRNA expression of human PBMC.Bisphosphonates may be used in the prevention and treatment of aseptic loosening of prosthesis by downregulating IL-6mRNA expression of human PBMC.
