摘    要: 目的考察SIPI在大鼠体内的代谢转化。方法采用液相色谱-质谱(LC-MSn)联用技术,检测在单剂量静脉注射给予SIPI后大鼠尿,粪及胆汁中的SIPI及代谢物。色谱柱为Diamonsil C18柱;流动相为甲醇-水-甲酸(40∶60∶0.5),流速为0.5mL·min-1;质谱仪离子源为电喷雾离子源(ESI),正离子方式检测。代谢物经LC-MSn方法分离和分析,通过质谱和色谱行为推测其结构。结果在大鼠尿样中共检测到8种代谢物,在大鼠粪样中共检测到4种代谢物,在大鼠胆汁样品中共检测到3种代谢物。结论SIPI在大鼠体内广泛代谢,形成多种代谢产物。

Abstract:OBJECTIVE To investigate the metabolic profile of SIPI in rats.METHODS Liquid chromatography-tandem mass spectrometry (LC-MSn) was used for the separation and analysis of SIPI and its metabolites in rats urine, feces and bile after an intravenous dose of SIPI. The metabolites were separated using a mobile phase consisting of methanol-water-formic acid (40∶60∶0.5) at 0.5 mL·min-1 with a Diamonsil C18 column. The instrument was operated in positive electrospray ionization (ESI) mode. The metabolites were identified by comparisons of their mass spectra and LC behavior.RESULTS Totally,8 metabolites were detected in rats urine, 4 metabolites were detected in rats feces and 3 metabolites were detected in rats bile.CONCLUSION SIPI underwent extensive metabolic pathways in rats.
