肾阴虚抑郁症大鼠模型的建立及评价

目的建立肾阴虚抑郁症大鼠模型。方法将50只大鼠随机分为正常组、单纯应激组、温燥组、甲状腺素组及激怒组;采用慢性温和不可预见性慢性应激建立抑郁症"病"模的同时,正常组、单纯应激组灌胃生理盐水,甲状腺素组灌胃甲状腺素,温燥组灌胃温燥药;激怒组采用长期激怒法并灌胃生理盐水。使大鼠在抑郁的基础上表现出一定的阴虚"证,"以大鼠一般状态、敞箱试验、液体消耗试验、血清雌二醇(E2)、睾酮(T)及血浆环腺苷酸(cAMP)、环鸟苷酸(cGMP)作为检测指标,对各模型进行比较。结果与正常组比较,各模型组的体质量、水平得分、垂直得分、中央格停留时间、糖水消耗、总液体消耗各指标测值减少,温燥组、甲状腺素组及激怒组普遍出现活动减少,大便干燥,毛湿无光泽,易惊怒以及血浆cAMP含量升高,其中甲状腺素组和激怒组大鼠血清E2、E2/T比值显著升高。结论甲状腺素合并应激抑郁模型所致的肾阴虚抑郁,更符合临床实际,简单易行,可作为肾阴虚抑郁症造模方法之一。

Establishment and evaluation on experimental model of kidney-yin deficiency and depression syndrome

Objective To establish a better experimental model of kidney-yin deficiency and depression syndrome.Methods All 50 rats were divided into normal,stress only,warm-dryness,thyroxine and irritation groups;all the rats were mildly and chronically irritated to establish the models of depression syndrome.The rats was fed with normal saline in the normal and stress only groups,with thyroxine in thyroxine group and warm-dry medicine in warm-dryness group,and the rats in irritation group was irritated in long-term and fed with normal saline,which could cause rats in a depression state with kidney deficient syndrome.And all the general conditions,open-field test,fluid consumption,concentration of cAMP and cGMP in plasma,and concentration of E2,T in serum were detected and compared.Results Compared with normoal group,the targets in all groups,such as body weight,horizontal scores,vertical scores,stopping time in center,sugar water consumption and total body fluid consumption,were decreased,the motions of rats in thyroxine,warm-dryness and irritation groups were lowered with dry stool,easy irritation and increased serum cAMP concentration,in which,the ratio of serum E2,E2/T in thyroxine group and irritation group was increased obviously.Conclusion The model of kidney yin deficiency and depression syndrome,induced by feeding thyroxine and irritated depression,is simple and prepared easily,which can be the first selection for the modeling of kidney yin deficiency and depression syndrome.
