摘    要:目的建立以高效液相色谱法测定人尿中酮洛酸浓度的方法。方法色谱柱为Waters Sunfire C18,流动相为0.02mol·L-1磷酸二氢钾溶液-乙腈(70∶30,用磷酸调节pH值至4.5),检测波长为315nm,柱温为35℃,流速为1mL.min-1,进样量为20μL,保留时间为9.1min。结果人尿中酮洛酸浓度在0.1~10.0μg·mL-1范围内线性关系良好,日内和日间RSD<10%,最低定量限为0.1μg·mL-1。结论本法简便、快捷、灵敏,适用于酮洛酸人体药动学研究。

Abstract:OBJECTIVE:To establish a HPLC method for the determination of ketorolac in human urine.METHODS:The chromatographic column was Waters Sunfire C18,the mobile phase consisted of 0.02mol·L-1 potassium dihydrogen phosphate-methyl cyanide(70∶30,adjusted to pH4.5 with phosphoric acid).The detection wavelength was 315nm;column temperature was 35℃;the flow rate was 1mL·min-1 The sample size was 20μL and retention time was 9.1min.RESULTS:The linear range of ketorolac concentration in human urine was 0.1~10.0μg·mL-1,The inter-day RSD and intra-day RSD were less than 10%.The minimum quantity limitation was 0.1μg·mL-1.CONCLUSION:This method is a sensitive,convenient and feasible for ketorolac detection.
