沙利度胺对内毒素刺激大鼠腹腔巨噬细胞分泌TNF-α和IL-6的影响

目的探讨沙利度胺对内毒素刺激大鼠腹腔巨噬细胞分泌TNF-α和IL-6的影响。方法分离、培养Wistar大鼠腹腔巨噬细胞，贴壁后的腹腔巨噬细胞分为空白对照组、内毒素（LPS）组、沙利度胺处理组、沙利度胺对照组。空白对照组不予任何处理；沙利度胺处理组提前2h加入不同浓度的沙利度胺，2h后加入内毒素；LPS组加入LPS，沙利度胺对照组仅加入沙利度胺80μg／mL。细胞培养4h后取上清液测TNF-α和IL-6浓度。结果空白对照组大鼠腹腔巨噬细胞上清TNF-α和IL-6含量均很低，沙利度胺对照组与空白组无显著性差异。大鼠腹腔巨噬细胞经LPS诱导后，细胞上清中TNF-α及IL-6含量显著增高，提前2h沙利度胺预处理后，TNF-α和IL-6的释放明显受到抑制，抑制作用随沙利度胺预处理浓度的增大而增强。结论沙利度胺能抑制大鼠巨噬细胞对TNF-α和IL-6等促炎细胞因子的释放，可能是其对抗脓毒症的机制之一。

Effects of Thalidomide on LPS Reduced TNF-α and IL-6 Release in Peritoneal Macrophages of Rat

Objective To investigate the effects of thalidomide on LPS reduced TNF-α and IL-6 release in peritoneal macrophages of rat. Methods Peritoneal maerophages of rat were separated and cultured. The cells were treated with lipopolysaccharide（LPS）. Thalidomide or LPS＋thalidomide respectively, and then incubated for 4 hours at 37℃ and 5% CO2. The supernatants of the cells were collected to detect the concentration of tumor necrosis factor-α （TNF-α）and interleukin-6 （IL-6）. Results In vitro experiment in peritoneal macrophages of rat showed that thalidomide obviously inhibited LPS-induced TNF-α and IL-6 release. The effect of inhibition was accentuated with thalidomide concentration increasing. Conclusion Thalidomide can evidently inhibit LPS-induced release of TNF-α and IL-6 from peritoneal macrophages of rat, indicating that it can suppress the inflammatory response caused by LPS.
