清脑益智方药血清在缺氧培养条件下对人胚大脑神经细胞的影响

摘    要:目的 :探讨清脑益智方药血清在缺氧条件下 ,对体外培养人胚脑神经细胞活性、自由基清除及细胞超微结构保护方面等作用。方法 :在缺氧条件下 (95 %N2 5 %CO2 ,12 0min) ,造成胚脑神经细胞损伤模型 ,应用清脑益智方药处理人血清 ,以牛磺酸为对照组进行比较性研究。结果 :清脑益智方药血清对缺氧损伤的神经细胞培养液中异常增高的LDH释出量有显著的降低作用 ,提高了SOD活力和NSE活性 ,降低LPO含量 ,有保护神经细胞的超微结构等作用。结论 :表明清脑益智方药人血清在缺氧培养条件下 ,对人胚大脑神经细胞有一定的保护作用。

Abstract:In this research,Qing Nao Yi Zhi Fang treated serum(QNYZs)has been developed as a drug to be used for the prevention and treatment of vascular dementia.However,the mechanisms by which this drug affects vascular dementia remain unknown.We examined the effects of QNYZ serum under oxygen deprivation condition (95%N 2 5%CO 2,120min) on human fetal cerebral neuronal cells in primary culture.On hypoxia condition,it leads to a decrease in the activies of cholinesterase(ChE),superoxide dismutase(SOD),NSE(Neuron specific enolase) and an increase in lactate dehydrogenase(LDH) release,lipid peroxidase(LPO).QNYZ serum could increase the activity of SOD,NSE (Neuron specific enolase),ChE(cholinesterase),and can decrease content of LPO,LDH release,and could protect the neuron form the injury of oxygen deprivation.Therefore,we consider that QNYZs has protective effects against oxygen deprivation condition in neuronal cells during ischemic brain injury.
