摘    要:目的研究微小RNA(microRNA,简称miRNA)与心肌细胞氧化应激的关系,作为探讨缺血再灌注损伤分子病理生理的基础。方法培养中的H9c2(2-1)心肌细胞以过氧化氢处理产生氧化应激,然后收集心肌细胞提取miRNA。选取miR-210和miR-24进行逆转录,再进行定量实时聚合酶链式反应。结果心肌细胞以过氧化氢处理2h后,与对照相比,miR-210显著降低至对照水平的47.07%(P=0.013),而miR-24的水平则大幅升高,达到了对照水平的344.48%(P=0.002)。结论 miRNA与心肌细胞氧化应激反应具有明显的相关,提示miRNA可能在心脏缺血再灌注损伤的病理损害和创伤修复中具有重要作用。

Abstract:Objective To investigate the relationship of microRNAs(briefly miRNA) and the oxidative stress in myocardial cells,as a basis for probing the underlying molecular pathophysiology of ischemia-reperfusion.Methods H9c2(2-1) myocardial cells in culture were exposed to hydrogen peroxide to induce cellular oxidative stress,followed by cell harvesting and miRNA extraction.miR-210 and miR-24 were reverse transcribed and then entered quantitative real-time PCR.Results 2h treatment of myocardial cells with hydrogen peroxide obviously decreased the cellular levels of miR-210 to 47.07%(P=0.013) of control levels while drastically increased miR-24 to 344.48%(P=0.002) of control ones.Conclusion There exists a clear connection between miRNAs and the oxidative stress of myocardial cells,suggesting miRNA may play crucial roles in the myocardial damage and recovery in cardiac ischemia-reperfusion.
