雷公藤多甙对大鼠局灶性脑缺血-再灌注损伤脑组织中NO水平、NOS活性的影响

摘    要:目的:研究大鼠局灶性脑缺血-再灌注损伤脑组织中一氧化氮(NO)水平,一氧化氮合酶(NOS)活性变化及雷公藤多甙(GTW)对其影响,方法:将100只Wistar大鼠分成假手术组,手术组,生理盐水治疗组和GTW治疗组,用线检法建立大鼠脑缺血-再灌注动物模型,用硝酸银还原法测定4例大鼠脑组织缺血1h再灌注15min,1h,6h,12h,24h,NO含量和NOS活性。结果:与手术组及生理盐水治疗组相比,各时间点GTW治疗组大鼠脑组织NOS活性降低,NO含量减少(P<0.05)。结论:GTW对大鼠局灶性脑缺血-再灌注损伤有保护作用。

Abstract:Aim:To study the changes of NOS and NO in tissues of focal cerebral isehemia-reperfusion injury in rats and the effects of multi-glycosides of TWHF (GTW) on them. Methods; A total of 100 Wistar rats were allocated into 4 groups: the sham-operation group, the operation group, the group treated by normal saline and the group treated by GTW. Cerebral isehemia-reperfusion injury model was established with suture method. NO and NOS were determined with method of silver nitrate deoxidization after 1h ischemia and reperfusion 15 mins, 1 h, 6 h, 12 h,and 24 h. Results:Compared to the operation group and the group treated by normal saline,at varied moments,the activity of NOS and the content of NO in group treated by GTW decreased (P <0. 05). Conclusion; GTW has protective effect on focal cerebral ischemia-reperfu-sion injury in rats.
