亚低温对心肺复苏后大鼠脑组织AQP4表达的影响

摘    要:目的 探讨亚低温对心肺复苏后大鼠脑组织AQP4表达的影响。方法 42只雄性SD大鼠随机分为3组:假手术组(6只),常温复苏组及亚低温组,后2组再分为3个亚组,即自主循环恢复(ROSC)后1、3、6h组,每个亚组均为6只大鼠,亚低温组复苏后即予亚低温干预。应用免疫组化法测脑组织AQP4表达.干湿重法测脑含水量。结果 亚低温组ROSC后1小时BWC与同一观察时点复苏组比较。无显著性差异。但AQP4表达水平下降已有显著性差异(P〈0.05)。亚低温组ROSC后3h和6h,BWC和AQP4表达水平与相同观察时点复苏组比较,均明显降低,差异性显著(P〈0.05)。结论 亚低温可减轻心肺复苏后脑水肿,其机制可能为:亚低温使大鼠脑组织AQP4表达下调。

Abstract:Objective To study the effects of mild hypothermia on expression of AQP4 in rats' brain tissue after cardiopulmonary resuscitation.Methods Forty-two male rats were randomly divided into three groups:sham-operated group(n=6),normal temperature group,and mild hypothermia group. The later two groups were again divided into ROSC1,3,6 hours subgroups(n=6).The expression of AQP4 in brain tissue was determined with semi-quantization immuno-histochemistry staining at ROSC1,3,6 hours,and the BWC was determined by brain ratio of wet to dry weight at the same time. Results The BWC and expression of AQP4 was significantly decreased at ROSC3 h, 6 h in mild hypothermia group compared with that of the same time in normal temperature group(P<0.05).Conclusions Mild hypothermia induced after CPR can down-regulate the expression of AQP4 in brain tissue and thus decrease brain edema,so mild hypothermia can be used as a useful treatment after CPR.
