摘    要:目的观测不同波长设置时,3种溶血程度对丙氨酸氨基转移酶(ALT)测定的影响。方法用Sysmex XT-1800i全自动血细胞分析仪测定血清中血红蛋白(Hb)浓度。应用Olympus AU2700全自动生化分析仪在波长设置于340nm和340/660nm时检测20例住院患者,在不溶血和溶血程度在Hb为2.0、5.0、8.0g/L时ALT指标的变化。结果应用配对设计差值均数t检验得出,波长340nm时,与无溶血比较,Hb-2.0g/L时差异无统计学意义(P〉O.05);Hb=5.0g/L时显著增高,差异有统计学意义(P〈O.05);Hb-8.0g/L时显著增高,差异有统计学意义(P〈0.01)。波长340/660nm时,与无溶血比较,Hb-2.0g/L时差异无统计学意义(P〉0.05);Hb-5.0g/L时差异无统计学意义(P〉0.05);Hb-8.0g/L时显著增高,差异有统计学意义(P〈0.01)。无溶血标本在两种波长设置检测结果差异无统计学意义(P〉0.05)。结论一定的溶血会影响ALT检测,不同波长设置抗干扰能力不同,340/660nm优选。

Abstract:Objective To observe the effects of three degrees of hemolysis in ALT determination with two different wavelengths. Methods To determine the concentration of hemoglobin in serum with Sysmex XT-1800i automatic blood analyzer, and determine the activities of ALT in serums, in which the concentration of hemoglobin has been determined(2.0 g/L,5.0 g/L,8.0 g/L) with Olympus AU2700(340 nm,340/660 nm). The serum were collected from 20 patients in the hospital. Results By means of partnership difference t-test ,when single wavelength is at 340 nm, there is no remarkable difference(P〉0.05) between serum without hemolysis and 2.0 g/L concentration in Hb ;but there is remarkable difference(P〈0.05) between serum without hemolysis and 5.0 g/L concentration in Hb ; There is remarkable difference(P〈0.01) between serum without hemolysis and 8.0 g/L concentration in Hb. By using double wavelengths(340/660 nm), there is no remarkable difference(P〉0.05) between serum without hemolysis and 2.0 g/L concentration in Hb ; There is no remarkable difference(P〉0.05) between serum without hemolysis and 5.0 g/L concentration in Hb ; There is remarkable difference(P〈0.01) between serum without hemolysis and 8.0 g/L concentration in Hb. There is no remarkable difference(P〉0.05) between single wavelength (340 nm) and double wavelengths(340/660 nm)if the serum is normal without hemolysis. Conclusion Control of influence on different degrees of hemolysis in ALT determination between single wavelength (340 nm) and double wavelengths (340/660 nm) is different. With single wavelength(340 nm), the exact results of ALT activity can be made if Hb interference is low. Proper wavelengths (340/660 nm) should be selected according to the increasing Hb interferences.
