Wistar大鼠代谢综合征动物模型的构建及其特征分析

摘    要:目的构建、评价高脂高盐膳食诱导的代谢综合征大鼠动物模型。方法8周龄雄性Wistar大鼠分为对照组和模型组(每组10只),分别给予普通膳食和高脂高盐膳食。然后每2-4周观测一次体重、血压和血糖,24周后进行腹腔葡萄糖耐量试验和正常葡萄糖高胰岛素钳夹试验,并观测血脂、内脏脂肪量和体长。结果模型组的体重、内脏脂肪、血压、胰岛素、甘油三脂均比对照组显著增加,并表现出显著的胰岛素抵抗。结论给大鼠高脂高盐膳食可诱导出符合或接近临床的代谢综合征。

Abstract:Objective To establish and evaluate a rat model of metabolic syndrome induced by high fat and salt diet.Methods Wistar rats,aged 8 weeks,were given routine diet(control) and high fat and salt diet(model),10 animals in each group.Then the body weights,blood pressure and blood glucose were measured every 2-4 weeks;24 weeks later,an intraperitoneal glucose tolerance test and a hyperinsulinemic euglycemic clamp test were performed,and the plasma lipids,the amount of fat around visceral organs and body length were measured.Results In the model group,(i) the body weight,visceral fat,blood pressure,insulin and triglycerol levels were all significantly increased compared to the control,and(ii) significant insulin resistance occurred.Conclusions High fat and salt diet given to rats could induce a metabolic syndrome,which corresponds or is analogous to the clinical metabolic syndrome.
