摘    要:目的:考察用分光光度法测定灵芝中总三萜含量的可行性。方法:采用正交试验法和单因素试验法。结果:不同测定条件,测定的结果差异较大,5%香草醛-冰醋酸的用量以0.2mL为宜,高氯酸的用量以0.8mL为宜,水浴温度以60~65℃为宜、水浴中加热时间以10~15min为宜。结论:该方法测定的结果不一定能反映灵芝中三萜的真实含量,若一定采用该方法测定灵芝中的总三萜含量,应严格控制检测条件,特别是水浴温度和水浴中加热时间。

Abstract:Objective:To study on the feasibility of using spectrophotometry to determine the total content of triterpene in ganoderma. Methods:Orthogonal and single-factor tests were carried out. Results:Under different conditions,tests with different results appeared. The appropriate test conditions were:0.2 mL of 5% vanillin and glacial acetic acid,0.8 mL of perchloric acid,60 ~ 65 ℃ in water bath and heating time of 10 ~ 15 min. Conclusion: This method maybe cannot truly show the total content of triterpene in Ganoderma. If this method is to be employed,the test conditions should be strictly controlled,particularly the water bath temperature and the heating time.
