烧伤膏对烫伤小鼠皮肤细胞自由基代谢和DNA损伤的影响

摘    要:目的:观察烧伤膏对烫伤小鼠皮肤组织细胞自由基代谢和DNA损伤的影响,探讨烧伤膏治疗烫伤皮肤的作用机制。方法:将50只小鼠随机分为5组:正常组、模型组,烧伤膏高、中、低剂量组。除正常组和模型组外,烧伤膏实验组小鼠每天外涂烧伤膏治疗,连用15天。治疗结束后,检测各组小鼠皮肤细胞中超氧化物歧化酶(SOD)、过氧化脂质(LPO)和各种自由基浓度,采用单细胞电泳技术检测DNA慧星细胞数和慧星平均尾长。结果:模型组小鼠皮肤细胞中SOD活力明显低于正常组(P<0.01),LPO和自由基浓度均比正常组明显升高(P<0.01)。与正常组相比,模型组小鼠皮肤细胞中慧星细胞数显著上升,DNA慧星平均拖尾明显加长,而应用烧伤膏治疗后,各项实验指标均明显得到改善。结论:烧伤膏可显著提高烫伤小鼠皮肤细胞中抗氧化酶活性,减少脂质过氧化产物和自由基的生成,对烫伤皮肤中DNA损伤有明显的保护效应。

Abstract:Objective:To study the effects of burn ointment on free radical metabolism and the DNA damage of skin cells of scalded mice,and mechanism of burn ointment for healing scalded skin.Methods:Fifty mice were divided into 5 groups randomly(i.e.control group,model group and groups with high,middle or low dose of burn ointment).Burn ointment was applied to the mice in experimental groups for 15 days except the control and model groups.SOD,LPO and free radical concentration of the skin cells of scalded mice were detected.Single cell gel electrophoresis(SCGE) was used to determine the number of comet cells and the comet tail length of DNA.Results:The SOD activities of the model group were apparently lower than that of control group(P 0.01).LPO and free radical concentration of the model group were all notably higher than that of control group(P 0.01).Compared with the control group,comet cells in skin of scalded mice of model group obviously increased while comet tail shortened.After using burn ointment,each indicator in the experiment was significantly improved.Conclusion:The burn ointment can significantly increase the activity of antioxidases and decrease the generation of LPO and free radical,which demonstrated the effect of burn ointment for the protection of DNA damage of skin cells of scalded mice.
