摘    要:目的建立尿毒清颗粒中黄芪甲苷的含量测定方法。方法采用高效液相色谱-蒸发光龊射检测器(HPLC—ELSD)法,色谱柱为Thermo Hypersil ODS-2柱(150mm×4.6mm,5μm),流动相为乙腈-水(30:70),柱温为30℃,蒸发光散射检测器漂移管温度105℃,载气流量2.8L/min。结果黄芪甲苷进样量在0.6276~3.9225μg范围内与峰面积呈良好的线性关系(r=0.9993),平均回收率为97.45%.RSD为3.44%(n=6)。结论HPLC—ELSD法准确、重现性好,可作为尿毒清颗粒的质量控制方法。

Abstract:Objective To establish a HPLC- ELSD determination method for astragaloside in Niaoduqing Granules. Methods The separation was performed on Thermo ODS- 2 Hypersil column(150 mm ×4.6 mm, 5 μm), the mobile phase was acetonitrile- water(30 : 70), the flow rate was 1 mL/min, the column temperature was 30 ℃. The evaporative light scattering detector(ELSD) as the detector, the temperature of the drift tube was set at 105 ℃, and the flow of gas was set at 2. 8 L/min. Results The linear range of astragaloside was 0. 627 6-3. 922 5 μg (r=0. 999 3). The recovery rate was 97.45%, RSD was 3.44% (n =6). Conclusion The method is accurate, precise, repeatable and suitable for the determination of astragaloside in Niaoduqing Granules.
