| 摘 要： | 目的:研究川楝子水提物对肺癌A549细胞的作用,初步探讨其作用机制。方法:采用MTT法检测川棟子水提物对A549细胞的增殖抑制率,同时观察细胞形态变化;qPCR法检测川楝子水提物作用后p53、Bax、Bcl-2、Bad和Bcl-xl等在mRNA水平的相对变化。结果:川楝子水提物可抑制肺癌A549细胞的增殖,其抑制率与作用时间和作用浓度呈正相关;实验组较对照组p53的mRNA表达量上调,Bax和Bad的mRNA表达量上调,Bcl-2和Bcl-xl的mRNA表达量下调。结论:川楝子水提物可抑制A549细胞增殖,可能与其激活P53基因和Bcl-2家族诱导细胞凋亡有关。

| Abstract: | Objective:To investigate the effect of Toosendan aqueous extract on human lung cancer cell line A549 cells.Methods:This study was divided into experimental group (concentration of 200,150,100,50μg/ml) and control group.The inhibitory rates of A549 cells effected by Toosendan aqueous extract were detected using MTT assay,and the changes of cell morphology were observed by microscope.The mRNAs expression of of p53,Bax,Bcl-2,Bad and Bcl-xl were detected using quantitative real-time PCR assay.Results:Proliferation of A549 cell was inhibited by Toosendan aqueous extract,and inhibition rates were related with time and the extract concentration in MTT assay.Compared with the control group,expression of P53,Bax and Bad mRNA increased,and expression of Bcl-2 and Bcl-xl mRNAs decreased in the experimental group.Conclusion:Toosendan aqueous extract may inhibit proliferation of A549 cells via activation of P53 gene and mitochondrial apoptotic pathway. |
