摘    要:目的:研究MTS1/p16基因在人类恶性肿瘤组织中的变化.方法:采用Southern杂交和多重PCR技术,分析了68例原发肿瘤组织标本,包括非小细胞肺癌31例、食管癌15例、胃癌13例、乳腺癌9例.结果:PCR检测癌旁组织未见p16基因缺失,而不同肿瘤组织标本的p16基因缺失差别很大,总缺失率为13.2%(9/68).Southern杂交检测p16基因的缺失率为17.7%(12/68).结论:MTS1/P16基因缺失在人类恶性肿瘤发生发展中起重要作用,是重要的多肿瘤抑制基因.

Abstract:Objective: To investigate the alteration of MTSl/pl6 gene in human primary cancers. Method:PCR and Southern blotting were performed on 68 fresh tumor specimens from cancer patients, including 31 lung cancers, 15 esophageal cancers, 13 gastric cancers and 9 breast cancers. Results: The results showed that no deletion was detected in 68 tumor - adjacent tissue samples. However, Diffrerent rate of gene deletion in different tumors was detected by PCR in 68 fresh tumor tissues.Total deletion rate of p16 was 13.2% (9/68) . It was confirmed by Southern blotting analysis with p16 cDNA probe. Conclusion: These date suggest that MTSl/pl6 gene alteration plays a critical role in the development of some cancers. p16 is an important suppressor gene of many tumors.
