银染端粒重复序列扩增法检测端粒酶活性及其临床应用

摘    要:目的 建立银染端粒重复序列扩增法(TRAP)测定肿瘤细胞的端粒酶活性.方法:将银染与TRAP相结合,测定了人肿瘤细胞株及穿刺细胞标本的端粒酶活性.结果:人肿瘤细胞株端粒酶活性为阳性,35例病理诊断为恶性肿瘤的穿刺标本中,33例端粒酶活性阳性,12例良性病变均为阴性.结论:银染TRAP法是特异,敏感及快速的端粒酶活性的检测方法,提示该方法可成为恶性肿瘤的诊断指标之一.

Abstract:Objective: To detect the telomerase activity of human tumor cells by silver staining telomeric repeat amplicatioin protocol (TRAP). Methods: Combining silver stain with TRAP, the telomerase activity of human tumor cells and fine - needle - aspri-ated samples were detected. Remits:The human tumor cells were positive for the telomerase activity. Among 35 cancerous fine - nee-dle- aspriated samples, 33 were positive.Telomerase activity was not detected in 12 negative controls.Conclusions: Silver staining telomeric repeat amplication protocol is specific, sensitive and fast assay for telomerase activity, and it could be one of the marks in cnacer.
