兔激素性股骨头坏死模型的研究

摘    要:目的研究内毒素脂多糖(LPS)+地塞米松注射液诱导兔股骨头坏死的实验效果。方法取新西兰大白兔36只随机分为模型组(n=21)和对照组(n=15)。模型组连续2 d每天经耳缘静脉注射内毒素脂多糖10μg/kg,再连续3 d每天臀部肌肉注射地塞米松注射液25 mg/kg;对照组注射同等剂量的生理盐水。结果 4周后,X线显示:模型组兔关节间隙增宽,密度增大,关节软骨下骨密度增高,股骨头变平,骨小梁模糊,软骨下骨与骨松质界限不清,在股骨头内出现斑块状高密度区域,股骨颈变短粗。双能量X线骨密度测量仪进行股骨头局部骨密度测量,兔股骨头骨密度检测发现模型组股骨头骨密度、骨矿物质含量,均显著低于对照组(P均<0.05)。组织学切片见模型组骨细胞陷窝空疏,脂肪细胞增多,部分血管栓塞,其中存活动物的骨坏死率和骨陷窝率均明显高于对照组(P均<0.05)。结论地塞米松联合脂多糖可有效建立激素性股骨头坏死模型。

Abstract:Objective It is to study the effects of caput femoris necrosis model of rabbits induced by lipopolysaccharide (LPS) and dexamethasone injection. Methods 36 New Zealand white rabbits were randomly divided into the model group (n = 21 ) and the control group (n = 15). The Model group was given lipopolysaceharide ( 10μg/kg) by the ear vein for two days, and given dexamethasone (25 mg/kg) by the hip muscles for three days. The control group was given the same dose of saline. Results After 4 weeks, X - ray showed : Rabbit articular of the model group were Gap widened,density increased. The density of joint subchondral bone increased. The caput femoris flattened out. Trabecular bone fuzzyed. The boundary of subchondral bone and Cancellous bone was unclear. Patchy high-density areas was found in caput femoris, femoral neck became short and thick. Took the Regional bone mineral density measurement of caput fcmoris by Dual-energy X - ray absorptiometry measuring instrument. Density detection of rabbits caput femoris showed bone mineral density and bone mineral content of caput femoris of the model group were significantly lower than that of the control group ( P 〈 O. 05 ). In the histological sections, bone cell lacu- nae of the model group was empty and sparse,fat cells increased. Some blood embolized. The rates of bone necrosis and bone lacuna rate of surviving animals were significantly higher than that of the control group (P 〈 O. 05 ). Conclusion Dexametha- sone combined with LPS can effectively establish caput femoris necrosis model of hormone.
