甘草活性成分对白癜风豚鼠模型的治疗作用

摘    要:目的:探讨甘草活性成分对白癜风豚鼠模型的治疗作用,阐明其对黑素生成的促进机理,为其应用于临床提供实验依据。方法:用水提法及大孔树脂分离纯化从甘草中提取活性成分,用过氧化氢复制白癜风豚鼠模型,造模成功,豚鼠随机分为模型组及低、中、高剂量治疗组,设正常对照组,经过甘草活性成分20、40和80 mg·kg-1 3个剂量治疗后,分别与模型组比较,观察用药部位大体改变、治疗总有效率、黑素细胞硫酸亚铁 Lillie特殊染色及HMB45免疫组织化学染色的组织学改变。结果:肉眼观察,治疗组肤色变浅程度弱于模型组,40和80 mg·kg-1组治疗总有效率高于模型组(P<0.05或P<0.01),且总有效率随着治疗剂量增加而增高;组织学观察,40与80 mg·kg-1治疗组豚鼠皮肤表皮基底层、棘层黑色素及毛囊黑色素增多,有黑色素的毛囊数增多,治疗组有黑色素的毛囊数多于模型组(P<0.01);HMB45免疫组织化学染色切片,40与80 mg·kg-1治疗组灰度值低于模型组(P<0.05或P<0.01),且平均灰度值随治疗剂量增大呈现递减趋势,表明40与80 mg·kg-1治疗组HMB45的表达量高于模型组且存在剂量依赖关系。结论:甘草活性成分可以促进有活性的黑素细胞增生,从而促进黑素的生成,对白癜风豚鼠模型有一定的治疗作用,提示甘草活性成分可应用于白癜风的临床治疗。

Abstract:Objective To investigate the therapeutic effect of glycyrrhiza active substance on guinea pig model of vitiligo,and to clarify its mechanism on promoting synthesis of melanin,and to provide the basis for its clinical application.Methods Guinea pigs decolorized by H2O2 were used as experimental models,and normal control group was set up.After treatment of glycyrrhiza active substance in three doses groups(20,40,and 80 mg·kg-1),the therapeutic effect was observed by visual observation,macrography,histological examination as well as immunohistochemical study.Results The significant differences of skin color among control group,model group and treatment groups were observed by macrography.The total effective rates in 40 or 80 mg·kg-1 groups were higher than that in control group(P<0.05 or P<0.01),and increased with the dose of glycyrrhiza active substance.Histological examination showed an evidently higher of melanin in basal cells,stratum spinosum and hair follicles in 40 or 80 mg·kg-1 treatment groups compared with model group(P<0.01).Immunohistochemical results showed the expressions of HMB45 were higher in 40 or 80 mg·kg-1 groups than that in model group(P<0.05 or P<0.01),and had dose-dependent relationship with glycyrrhiza active substance.Conclusion Glycyrrhiza active substance could increase the expression of HMB45 and promote synthesis of melanin,it shows certain efficacy in treatment of vitiligo,and it can be used in treating vitiligo in clinic.
