子宫内膜癌组织的FNDC10水平及临床意义

目的探讨子宫内膜癌(EC)组织的Ⅲ型纤维连接蛋白域蛋白10(FNDC10)水平及临床意义。方法采用GEPIA分析癌症基因组图谱(TCGA)数据库中EC组织的FNDC10水平,实时定量PCR(QPCR)检测97例EC组织、67例不典型增生子宫内膜组织和42例正常子宫内膜组织的FNDC10水平,绘制受试者工作特征(ROC)曲线以评估组织FNDC10水平诊断EC的效能和最佳阈值,进一步分析FNDC10水平与EC临床病理特征的关系,通过Kaplan-Meier Plotter数据库在线分析FNDC10水平与EC预后的关系。结果GEPIA在线分析显示,174例EC组织的FNDC10水平高于13例正常组织(P<0.05);QPCR结果显示,97例EC组织、67例不典型增生子宫内膜组织和42例正常子宫内膜组织的FNDC10水平依次为2.788±1.253、1.874±0.759和1.041±0.541,EC组织的FNDC10水平高于其余子宫内膜组织,而不典型增生子宫内膜组织的FNDC10水平也高于正常子宫内膜组织,以上差异有统计学意义(P<0.05);ROC曲线分析显示,组织FNDC10水平诊断EC的曲线下面积为0.900(95%CI:0.850~0.951,P<0.05),当以1.911为最佳诊断阈值时对应的约登指数最大,敏感度和特异度分别为73.20%和97.62%。FNDC10水平与年龄和病理类型无关(P>0.05),而与组织学分级、FIGO分期、肌层浸润深度和淋巴结转移有关(P<0.05)。Kaplan-Meier Plotter在线分析显示EC组织FNDC10水平与总生存期(OS)有关,其中高水平的中位OS为36.03个月,短于低水平的103.73个月(HR=1.98,95%CI:1.31~3.00)。结论EC组织的FNDC10水平升高且促进了该恶性肿瘤的病情进展,与预后有关,其水平有助于判断EC的组织学分级、FIGO分期、肌层浸润深度和淋巴结转移情况,在EC诊治和预后判断上有重要价值。

Level and clinical significance of FNDC10 in endometrial carcinoma tissues

Objective To investigate the level and clinical significance of fibronectin type III domain containing 10(FNDC10)in endometrial carcinoma(EC)tissues.Methods GEPIA was used to analyze the FNDC10 level of EC tissues in TCGA database,and real-time quantitative PCR(QPCR)was applied to detect the FNDC10 levels of 97 cases of EC tissues,67 cases of atypical hyperplasia endometrial tissues and 42 cases of normal endometrial tissues.The receiver operating characteristic(ROC)curve was drawn to evaluate the efficacy and best diagnostic threshold of FNDC10 for EC.The relationship between FNDC10 level and clinicopathological characteristics of EC was further analyzed.The relationship between FNDC10 level and prognosis of EC was analyzed online by Kaplan-Meier Plotter database.Results GEPIA online analysis showed that the FNDC10 level of 174 EC tissues was higher than that of 13 normal tissues(P<0.05).Results of QPCR showed that the FNDC10 levels of 97 EC tissues,67 atypical hyperplasia endometrial tissues and 42 normal endometrial tissues were 2.788±1.253,1.874±0.759 and 1.041±0.541,respectively.The FNDC10 level of EC tissues was higher than that of other endometrial tissues(P<0.05).The FNDC10 level of atypical hyperplasia endometrial tissues was also higher than that in normal endometrial tissues(P<0.05).ROC curve analysis showed that the area under the curve of FNDC10 level in tissue was 0.900(95%CI:0.850-0.951,P<0.05).When 1.911 was chosen as the best diagnostic threshold,the corresponding Yoden index was the highest with the sensitivity of 73.20%and specificity of 97.62%.FNDC10 level was not related to age and pathological type(P>0.05),but related to histological grade,FIGO stage,depth of myometrial invasion and lymph node metastasis(P<0.05).Kaplan-Meier Plotter online analysis showed that the FNDC10 level of EC tissues was related to the overall survival(OS).The median OS of the high level was 36.03 months,shorter than 103.73 months of the low level(HR=1.98,95%CI:1.31-3.00).Conclusion The increased FNDC10 level promoted the progression of EC,which was related to the prognosis.The FNDC10 level was helpful to judge the histological grade,FIGO stage,the depth of myometrial invasion and lymph node metastasis of EC,and it was of great value in the diagnosis,treatment and prognosis prediction of EC.
