Rac1蛋白在人卵巢浆液性腺癌组织中的表达及其对卵巢癌细胞SKOV3迁移能力的影响

摘    要:目的探讨Rac 1蛋白在人卵巢浆液性腺癌组织中的表达情况及其对卵巢癌细胞SKOV3迁移能力的影响。方法采用蛋白质印迹法(Western blot)检测59例卵巢浆液性腺癌组织、31例卵巢良性浆液性囊腺瘤组织和3 5例卵巢正常组织中Rac 1蛋白的相对表达量。筛选出抑制Rac 1蛋白表达效果最佳的载体,构建稳定转染细胞株SKOV3-Racli;采用细胞划痕实验检测SKOV3-Racli细胞的迁移能力。结果卵巢浆液性腺癌组织中Racl蛋白的相对表达量高于卵巢良性浆液性囊腺瘤组织和卵巢正常组织(P<0.05)。与阴性质粒比较,1号片段质粒的抑制率为12.59%,2号片段质粒的抑制率为56.48%,3号片段质粒的抑制率为73.68%。与野生型SKOV3组相比,SKOV3-Racli细胞组SKOV3-Racli细胞的迁移能力下降(P <0.05)。结论 Rac1蛋白对卵巢浆液性腺癌细胞迁移能力的影响可能与下调卵巢浆液性腺癌组织中Rac 1蛋白的表达有关。

Abstract:Objective To investigate the expression of Racl protein in human ovarian serous adenocarcinoma tissues and its effect on migration of ovarian cancer cell SKOV3.Method Western blot was used to detect the relative expression of Racl protein in 59 samples with ovarian serous adenocarcinoma,31 samples with benign ovarian serous cystadenoma and 35 samples with normal ovarian tissues.The vector with the best inhibition effect on Racl protein expression was selected,SKOV3-Racli,a cell strain with stable Racl expression deletion,was constructed.The migration ability of SKOV3-Racli was detected by cell scratch test.Result The expression of Racl in ovarian serous adenocarcinoma was higher than that in ovarian benign serous cystadenoma and normal ovarian tissues(P<0.05).The inhibitory rate of fragment 1 to 3 was 12.59%,56.48% and 73.68% respectively compared with negative plasmid.The migration ability of SKOV3-Racli cell group was significantly decreased compared with wide-type SKOV3 group(P<0.05).Conclusion The effect of Rac1 protein on the migration of ovarian serous adenocarcinoma cells may be associated with down-regulation of Racl protein expression in ovarian serous adenocarcinoma tissues.
