脑缺血再灌流后p53基因表达与细胞凋亡间的关系

摘    要:用原位杂交、免疫组化及原位末端标记(TUNEL)的方法观察大鼠大脑中动脉闭塞2h后再通0~72h脑组织p53基因的表达与凋亡细胞的分布.结果发现缺血2h及再灌流1h即可见p53mRNA及其蛋白的表达和凋亡细胞,p53mRNA及其蛋白的表达高峰在缺血再灌流后24h,凋亡细胞数高峰在再灌流24h~48h.提示脑缺血再灌流后诱导p53mRNA及其蛋白的表达和细胞凋亡.

Abstract:By using in situ hybridization method, immunohistochemistry and TUNEL, P53 mRNA and protein expression was detected and the distribution of apoptosis cells was observed in rat brain after 2 h occlusion of cerebral middle artery followed by 0 to 72 h reperfusion It was found that the expression of P53 mRNA and protein was detectable and apoptic cells could be observed at 1 h reperfusion after ischemia for 2 h P53 mRNA and protein expression reached peak at 24 h and cell apoptosis reached peak at 24 to 48 h of ischemia reperfusion Ischemia reperfusion could induce P53 gene and protein expression and apoptosis of cells
