康复训练对脑梗死大鼠大脑皮质突触膜糖蛋白表达的影响

摘    要:目的 研究康复训练对脑梗死大鼠大脑皮质中突触膜糖蛋白 (synaptophysin ,p38)表达的影响。方法 大鼠脑梗死 2 4h后随机分为康复训练组和制动组 ,康复训练组每天进行水迷宫、转棒、滚笼训练 ,制动组网状笼内固定。在 2 4h和 1、2、3、4周时采用免疫组织化学和图像分析技术观察大脑皮质p38的表达。结果 正常大鼠大脑皮质中可见到点状的p38免疫阳性物质 ,梗死后 2 4h及第 1周时在梗死灶周围皮质及对侧相应皮质中免疫阳性产物的灰度值无明显变化 ,2、3、4周时 ,梗死大鼠较正常大鼠灰度值明显增加 ;康复训练组与制动组比较 ,第 3、4周时前者的增加更为明显。结论 脑损伤后神经组织中存在突触发生 ,并且康复训练可促进这种过程。

Effects of rehabilitation training on the expression of synaptophysin in the cortex of cerebral infracted rats
Abstract:Objective To study the effect of rehabilitation training on the expression of synaptophysin in the cortex of cerebral infarcted rats. Methods After 24h of cerebral infarction rats were divided into different groups randomly: rehabilitation group was given water maze, rotating bar and rolling cage exercises six times a week while controlled group was immobilized in cages. Immunohistochemistry and image analysis technique were used to detect the expression of synaptophysin at 24 h and 1,2,3,4 weeks after infarction respectively. Results Synaptophysin appeared as a finely punctate pattern in normal cortex of both sides. The grey level of synaptophysin immunoreactivity did not demonstrate no statistically significant changes at 24 h and 1 week after infarction but,howed significant increases at 2,3,4 weeks in the parietal cortex lateral to the infarction area and the contralateral analogous region. And the incremental degree of rehabilitation rats was more than that of the immobilization rats at 3,4 weeks. Conclusion These data support the occurrence of synaptogenesis in the neocortex after infarction, which could be enhanced by rehabilitation training.
