蜂贝化瘀胶囊对2型糖尿病大鼠FasL基因表达和T细胞凋亡的影响

目的观察蜂贝化瘀胶囊对2型糖尿病大鼠FasL基因表达和T细胞凋亡的影响,比较中药治疗前后肾脏和视网膜病理变化的改变,并探讨其作用机制。方法建立2型糖尿病大鼠模型,通过RT-PCR、DNA凝胶电泳、流式细胞仪等技术,观察蜂贝化瘀胶囊对糖尿病大鼠FasL基因表达、T细胞凋亡的影响及电镜观察肾脏、视网膜病理变化。结果糖尿病模型组大鼠FasL基因表达和T细胞凋亡率明显高于正常对照组(P<0.05),而中药治疗组以上指标均明显降低(P<0.05),与正常对照组比较无显著性差异(P>0.05);中药治疗组与西药治疗组相比较,T细胞凋亡率也明显降低(P<0.05);电镜结果显示,中药治疗组肾脏和视网膜病理变化较糖尿病模型组明显减轻。结论蜂贝化瘀胶囊可明显降低T细胞凋亡率,明显改善肾脏病理变化,其作用机理是通过控制功能基因FasL基因的表达,减少了T细胞凋亡,从而增强免疫功能来实现的。

Effect of Fengbei Huayu Capsule on FasL Gene Expression and T Cell Apoptosis of Type 2 Diabetic Rats

Objective To study the effect of Fengbei Huayu capsule on FasL gene expression and T cell apoptosis of type 2 diabetic rats, and to compare the pathological lesions of kidney and retina after treatment of traditional medicine. Methods To make the model of type 2 diabetic rats and determine the indexes of FasL gene expression and T cell apoptosis rate by RT-PCR, DNA gel electrophoresis methods. Results Compared the FasL gene expression and T cell apoptosis rate, TCM treatment group was significantly lower (P <0.05) than those in the DM group, with no significant differences with control group (P >0.05). Comparison of FasL gene express, TCM treatment group was significantly lower (P <0.05) than those in the western medicine treatment group. In the TCM treatment group, the renal and retina pathological lesions were all alleviated compared with those in the DM group. Conclusion Fengbei Huayu Capsule can significantly decrease T cell apoptosis and alleviate the renal and retina pathological lesions, suggesting that it has better effect on controlling related gene expression, decreasing T cell apoptosis and improving immune function of the body.
