摘    要:目的 我们用物理化学方法处理自制的天然生物衍生骨支架材料一犬脱抗原松质骨(AECB),并复合具有成骨活性的骨形态发生蛋白(BMP)和具有成血管作用的碱性成纤维细胞生长因子(bFGF),制成复合型生物衍生组织工程骨。测其理化性质及力学强度,为组织工程研究提供可供选择的植骨材料。方法 将犬松质骨进行一系列理化处理,制成脱抗原松质骨,用化学方法分析其组成成分,用扫描电镜观察其形貌特征,用X线能谱仪测定其钙/磷比值,并对材料的力学性能进行分析测定,然后用物理方法在材料上复合BMP和bFGF,再用扫描电镜观察其复合情况。结果 犬脱抗原松质骨脱钙及有机质洗脱行之有效,保持原骨天然网状孔隙系统及一定力学强度,并且BMP和hFGF能与其有效结合。结论 犬脱抗原松质骨/BMP/bFGF有望为骨缺损、骨坏死的治疗开辟一条新途径,尚需要检测其免疫原性及成骨、成血管能力。

Abstract:Objective To detect the properties of composite derived material of canine antigen-extracted cancellous bone which was treated with different physical and chemical method, and was mixed with osteoinductive BMP and angiogenerable bFGF. Methods We made antigen-extracted cancellous bone from canine.Their morphological features, constitute components, ratios of calcium to phosphorus and mechanical properties were examined by scanning electron microscopy, X-rays diffraction analysis, mechanical assay and so on. At the same time, the morphological features of composite derived material were examined by scanning electron microscopy also. Results The calcium-extracted and organic-extracted process was available. The antigen-extracted cancellous bone possess natural network pore system. The BMP and bFGF were seen in the network pore system.Conclusion Canine antigen-extracted cancellous bone/BMP/bFGF would be a useful composite material in treating bone defect and osteonecrosis, but more inverstigations will be necessary to detect its immunogenicity,bone induction and angiogenesis.
