RRM1蛋白在非小细胞肺癌中的表达及临床意义

目的：研究核苷酸还原酶M1亚基（ribonucleotide reductase subunit M1,RRM1）蛋白在可手术的非小细胞肺癌（non-small-cell lung cancer,NSCLC）中的表达及RRM1蛋白表达阴性及阳性对不同方案辅助化疗无病生存期（dis-ease free survival,DFS）的影响。方法：免疫组化法回顾性检测51例Ⅰ~ⅢA期NSCLC根治术后标本RRM1蛋白的表达,分析其表达状况与患者临床特征之间的关系,并研究其对不同含铂二联方案辅助化疗患者DFS的影响。结果：（1）RRM1蛋白在不同的性别、年龄、吸烟状况、病理类型和病理分期患者中的表达差异均无统计学意义（P〉0.05）;（2）RRM1蛋白表达阴性时吉西他滨（gemcitabine,GEM）/铂类方案辅助化疗患者DFS优于非GEM/铂类方案（P〈0.05）;（3）COX多因素分析显示RRM1蛋白表达状况（P〈0.01）、病理分期（P〈0.01）为DFS的独立影响因素。结论：.RRM1蛋白是预测NSCLC患者GEM/铂类方案辅助化疗疗效的有价值指标。

Expression and clinical significance of RRM1 protein in non-small-cell pulmonary carcinoma

Objective： To investigate the expression of ribonucleotide reductase subunit M1（RRM1）protein in patients with resectable non-small-cell lung cancer（NSCLC）and the significance of adjuvant chemotherapy.Methods：51 patients with stage I to IIIA NSCLC undergone radical surgery were included in this study.The expression of RRM1 protein in tumor specimens was assayed by immunohistochemistry retrospectively.Correlations between the expression of RRM1 protein and the clinical features or the disease free survival（DFS） time after operation had been analysed.Results：（1） No differences in the expression of RRM1 protein had been found among patients with different gender,age,smoking status,pathology and stage （P0.05）.（2） When RRM1 protein expression was negative,adjuvant chemotherapy of GEM/platinum would lead to longer DFS time than others（P0.05）.（3） COX multivariate regression analysis showed that the expression of RRM1 protein （P 0.01）and pathological classification（P 0.01）were independent prognostic factors of DFS in postoperative patients.Conclusion： RRM1 protein is a valuable predictive factor for GEM/platinum in postoperative NSCLC patients.
