不同缺血时间对豚鼠离体心脏缺血/再灌注损伤的影响

摘    要:目的探讨不同缺血时间对豚鼠离体心脏缺血/再灌注损伤的影响。方法将23只豚鼠离体心脏随机分成4组:正常组(Control,n=6)、缺血30 min再灌60 min组(I30R60,n=6)、缺血40 min再灌60 min(I40R60,n=5)、缺血50 min再灌60 min组(I50R60,n=6),观察不同缺血组缺血前及再灌期间心率和冠脉流量,测定灌流液中乳酸脱氢酶(lactate dehydrogenase,LDH)和肌酸激酶(creatine ki-nase,CK)漏出量及组织中超氧化物歧化酶(superoxide dis-mutase,SOD)及丙二醛(malondialdehyde,MDA)含量,并以2,3,5-三苯基氯化四氮唑(2,3,5-triphenyltetrazolium chlorid,TTC)染色法测定梗死面积。结果与Control组相比,I30R60组各指标尚未全部发生明显变化;而I40R60和I50R60组心率和冠脉流量降低,梗死面积加大,灌流液中LDH和CK漏出值升高,组织中SOD和MDA值降低;上述所观察指标中,I50R60组变化更为明显。结论豚鼠离体心脏缺血/再灌注损伤受缺血时间的影响,豚鼠离体心脏缺血/再灌注损伤模型的建立以缺血50 min为宜,再灌时间可考虑限制于15~60 min之间。

Abstract:Aim To explore the effects of different ischemia and reperfusion time on ischemia reperfusion injury in heart of isolated guinea pig.Methods Twenty three isolated guinea pig hearts were divided into four groups: Control group(n=6),I30R60 group(n=6),I40R60 group(n=5) and I50R60 group(n=6) where hearts were subjected to global ischemia for 30 min,40 min and 50 min followed by reperfusion for 60 min.Then heart rate,coronary flow,lactate dehydrogenas(LDH) and creatine kinase(CK) in effluent,superoxide dismutase(SOD) and malondialdehyde(MDA) in heart tissues and infarct size(by TTC staining) were measured in different ischemic groups.Results Compared to control group,not all indexes in this experiment had changes in I30R60 group.While I40R60 and I50R60 groups had more reduction in HR,CF,larger infarct size,higher releases of LDH and CK in effluent,and lower SOD and MDA in heart tissues.Additionally,I50R60 group was different from control group in all indexes observed in this experiment significantly.Conclusions Myocardial ischemia reperfusion injury in isolated guinea pig heart is affected by ischemic time.50 min ischemic time is better for myocardial ischemia reperfusion injury model in isolated guinea pig heart,and 15 to 60 min reperfusion time can be considered.
