电离辐射对血管内皮细胞分泌与组织创伤相关细胞因子的影响

摘    要:目的探讨电离辐射对血管内皮细胞分泌血管内皮生长因子(VEGF)、碱性成纤维生长因子(bFGF)和转化生长因子β1(TGF-β1)的影响。方法对人脐静脉血管内皮细胞(HUVEC)-12以0、4、8、12Gy剂量60Coγ射线照射后培养12、24h,采用流式细胞仪检测细胞VEGF、bFGF和TGF-β1的表达。结果VEGF在4Gy照射后12h有较明显的增加,24h后逐渐趋于正常;bFGF在4Gy照射后12和24h都有明显的表达,但表达量随着辐射剂量的增加而减少;TGF-β1表达的增加呈剂量-时间依赖性。结论血管内皮细胞受电离辐射后VEGF、bFGF和TGF-β1的分泌均有增加,但三者的剂量-时间关系不一致,可能与细胞对辐射的反应和损伤抵抗作用不同有关。

Abstract:Objective To observe the effect of ionizing on the secretion of vasculare endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) and transforming growth factor-β1 (TGF-β1) in vascular endothelial cell. Methods The expression levels of VEGF, bFGF and TGF-β1 of human umbilical vein endothelial cell (HUVEC)-12 were detected by flow cytometry at 12- and 24-hours post-irration with 0, 4, 8 and 12 Gy ^60Co γ-rays. Results VEGF increased significantly at 12 hours post-irration with 4 Gy and gradually tended to be normal level at 24 hours. The expression of bFGF increased significantly at 12- and 24-hour with 4 Gy but decreased depending on the increasing of radiation dose. The expression of TGF-β1 increased with the radiation doses and times. Conclusion The secretion of VEGF, bFGF and TGF-β1 of vascular endothelial cells increased but the relationships with the radiation dosage and time were not consistent. That may be related to the cellular reaction and damage resistance to the ionizing radiation.
