基因重组人集落刺激因子对环磷酰胺处理小鼠造血干细胞的动员作用

摘    要:国产基因重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)和进口人粒细胞集落刺激因子(rhG-CSF),对C57小鼠环磷酰胺(CPA)应用所造成的骨髓抑制,无论在化疗药与造血因子同时应用的治疗组或造血因子先于化疗药应用的预防组,均显示出对造血干细胞明显的动员作用,与对照组相比有显著差异(P<0.01~P<0.001);rhGM-CSF升高白细胞(WBC)的作用稍次于rhG-CSF,但增加骨髓DNA含量作用稍强于rhG-CSF,无显著性差异(P>0.05)。同时,rhGM-CSF对骨髓干细胞中分化程度低的集落刺激作用强于分化程度高的小丛和大丛,rhG-CSF恰好相反。各测定指标显示:两种造血因子作用无显著差异。各药皆有一定的量效关系。

Abstract:It was observed that a China-made granulocyte macrophage colonystimulating factor(rhGM-CSF)and a granulocyte colony-stimulating factor ( rhG-CSF )made in Japan were able to mobilize the hematopoietic progenitor cells of mice treated by high-dose cyclophosphamide (CPA). Both rhCSFs injected subcutaneously in the CPA treated mice caused a neutrophilia in both therapeutic and prophylactic groups.The neutropenia of the therapeutic group was not more apparent than that of the prophylactic group, but the myeloblast DNA increased steadily in prophylactic group. The rhGM-CSF was more effective on the forming of the colony of immature haemapoietic progenitor cells,and in contrast, the rhG-CSF was more active in the forming of group, and the full mature ones. Effects of rhGM-CSF were practically identical,and showed a good tendency of dose dependant relationship.
