广西甜茶提取物的抗炎作用研究

摘    要:目的:研究广西甜茶提取物的抗炎作用。方法:观察广西甜茶提取物对巴豆油诱发小鼠急性耳肿胀和对血管通透性的抑制作用及角叉菜胶引起小鼠足跖肿胀的影响。利用Griess化学法检测小鼠腹腔巨噬细胞NO含量,RT-PCR检测一氧化氮合酶(iNOS)的表达。结果:广西甜茶提取物能显著抑制由巴豆油诱发的小鼠耳肿胀及血管通透性的增高,并对角叉菜胶引起的足跖肿胀也显示一定的抑制作用。广西甜茶提取物可抑制巨噬细胞NO生成和iNOSmRNA表达。结论:广西甜茶具有抗炎作用,其作用机制可能与抑制巨噬细胞NO生成和iNOSmRNA表达有关。

Abstract:OBJECTIVE: To investigate the anti-inflammatory effects of the extract of Rubus scuaivissimus cultivated in Guangxi. METHODS: The inhibition effects of the extract of R. scuaivissimus on vasopermeability and acute mice ear swelling induced by croton oil, and mice paw swelling induced by carrageenan were observed. The content of nitric oxide (NO) and the mRNA expression of iNOS in mice peritoneal macrophages were detected using Griess and RT-PCR assay. RESULTS: The extract of R. scuaivissimus cultivated in Guangxi can conspicuously inhibit the increase of vasopermeability and acute ear swelling induced by croton oil in mice,and mice paw swelling induced by carrageenan. Furthermore, R. scuaivissimus extract also showed depressant effect on the release of NO and genes expression of iNOS mRNA in mouse peritoneal macrophages. CONCLUSION: The extract of R. scuaivissimus cultivated in Guangxi has conspicuous anti-inflammatory effects. The mechanism of action may be associated with its inhibition effects on the release of NO and mRNA expression of iNOS.
