氯化两面针碱与兔血浆的蛋白结合率研究

摘    要:目的:测定氯化两面针碱与兔血浆蛋白的结合率。方法:通过平衡透析法结合高效液相色谱法测定氯化两面针碱与兔血浆蛋白的结合率。结果:氯化两面针碱与内标(氯霉素)完全分离,血浆中其它成分无干扰;其在0.03188~2.04000μg·mL-1浓度范围内与氯化两面针碱同氯霉素峰面积比线性关系良好;0.125、0.25、0.5μg·mL-1浓度样品的方法回收率均>95%,日内、日间精密度的RSD均<5%;3种药物浓度的血浆蛋白结合率分别为73.21%、72.83%、70.30%。结论:氯化两面针碱与兔血浆蛋白具有中等强度的蛋白结合率。

Abstract:OBJECTIVE : To determine the rabbit plasma protein binding rate of nitidine chloride. METHODS : The equilibrium dialysis combined with HPLC was used to determine the rabbit plasma protein binding rate of nitidine chloride. RESULTS : Nitidine chloride and the internal standard (chloromycein) were separated completely without interference. The linear range was 0.031 88 -2.040 00 μg·mL^-1. Its recovery rates were more than 95% at the concentrations of 0,125 μg·mL^-1, 0.25 μg·mL^-1, 0.5 μg·mL^-1. The RSD ofintra-day and inter-day both were less than 5%. The plasma protein binding rates of nitidine chloride with concentrations of 0.125 μg·mL^-1, 0.25 μg·mL^-1 and 0.5μg·mL^-1 were 73.21%, 72.83 %, 70,30 %, respectively. CONCLUSION: Nitidine chloride shows moderate rabbit plasma protein binding rate.
